GPC3 (Human Glypican-3) Immunoassay                                                                                                Catalog # B1507

 

The GPC3 Immunoassay is a sandwich-ELISA for the determination of human Glypican-3 protein concentrations in cell culture supernatants, cell lysates and human serum.  The assay employs two GPC3-specific monoclonal antibodies; one is immobilized to the surface of microtiter wells provided with the kit.  Samples or GPC3 standards are incubated for 2 hours, followed by a second 2 hour incubation with the secondary detector antibody.  Total time for the assay, including setup, wash and data collection, is approximately 6 hours.

 

Glypicans are a family of heparan sulfate proteoglycans that are linked to the exocytoplasmic surface of cell membranes by a glycosyl phosphatidylinositol (GPI) anchor.  Glypican-3 is an oncofetal antigen that has been found to be mutated in Simpson-Golabi-Behmel syndrome (SGBS).  This X-linked disorder is characterized by overgrowth and a broad spectrum of clinical manifestations.  In addition to its upregulation in certain cancers, GPC3 is downregulated in breast cancer, ovarian cancer and mesothelioma.  It is capable of activating the non-canonical Wnt pathway, and downregulation appears to be mediated by promoter methylation.  It is a useful immunohistochemical marker of early hepatocellular carcinoma.  It has been implicated in other types of cancer, including squamous cell carcinoma of the lung, melanoma and testicular germ cell tumors.
